The necessity of freezing and grinding in liquid nitrogen

In molecular biology experiments, animal muscle tissue, connective tissue, bone tissue, hair tissue, etc., and plant roots, stems, leaves, seeds, etc. need to be ground, and then their components, genes and proteins, and structure and function are analyzed. Research.

In order to make various tissue components not easily damaged or degraded during the grinding process, and to make the tissue hard and brittle, so that it is easy to grind, especially for the grinding, separation and extraction of ribonucleic acid (RNA) and deoxyribonucleic acid (DNA). . The biological material needs to be quickly frozen in liquid nitrogen to stop all biological reactions inside and outside the cell. At the same time, the biological material cells are completely frozen and hardened and brittle, so grinding can achieve a good cell breaking effect. substances are released.

Grind samples

Liquid nitrogen freeze grinding step

Aliquot tissue samples into appropriate centrifuge tubes.

Add an appropriate amount of lysis medium to the centrifuge tube

Screw the cap on the centrifuge and insert it into the freezer adapter.

Using the dedicated adapter holder, place the frozen adapter in its entirety in liquid nitrogen for 3-15 minutes.

Then use the special adapter fixture to place the frozen adapter as a whole, place it on the Gd16plus grinder, and tighten the fixed handle.

Set the grinding speed and time, start the Gd16plus grinder

After the grinder has stopped, remove the freezer adapter and remove the centrifuge tube.

Precautions

The temperature of liquid nitrogen at normal pressure is quite low. When using liquid nitrogen, extra care must be taken to avoid direct contact with the skin.

2. Placing and removing the cryo-adapter into and out of liquid nitrogen should be done with a special adapter clamp and anti-freeze gloves.
